Reveal the molecular mechanism behind the strong protective ability of aspirin

Now, in a new study, researchers from the Duke University Department of Medicine have identified a new mechanism of action for aspirin that seems to explain the many benefits of the drug. The relevant research results are published in the EBioMedicine journal entitled "Systems Pharmacogenomics Finds RUNX1 Is an Aspirin-Responsive Transcription Factor Linked to Cardiovascular Disease and Colon Cancer".
The researchers described how aspirin directly affects the function of a gene regulatory protein that not only affects platelet function but also inhibits colon tumors.
Dr. Deepak Voora, co-author of the paper and an assistant professor of applied genomics and precision medicine at Duke University, said, "This study identified a new mode of action for aspirin, which is unpredictable based on known pharmacological effects. Voora said that the ability of aspirin to reduce pain and dilute blood has long been attributed to its ability to block COX-1, an enzyme involved in inflammation and blood clotting.
“But COX-1 only partially explains how aspirin is good for cardiovascular health, but it has not been proven to be involved in cancer,†he said.
Instead, Voora and colleagues focused on the gene activity profile they call the aspirin response signature. Based on this response profile, they identified a network of genes associated with platelets and heart attacks.
"This method of using genomics data to comprehensively evaluate the mechanism of action of a drug -- as we are in this study," said Dr. Geoffrey Ginsburg, co-author of the paper and director of the Center for Applied Genomics and Precision Medicine at Duke University. The work of using aspirin is a change that can change the way drugs are developed and used in clinical treatment.
