The Azure Biosystems c-Series digital imaging family is the industry's leading product with a user-friendly interface and flexible applications. In this application, we showcased some new applications that can be completed on the Azure Biosystems c280, the newest member of the c-series family.
Introduction
The Azure Biosystems c280 digital imager is the ideal entry point for digital chemiluminescence imaging. However, its advantages do not stop there. C280 with dual wavelengths of 302 nm and 365 nm UV is used for imaging of ethidium bromide (EtBr) DNA gel; EPI blue LEDs are used for imaging of "safe" DNA dye SYBR Green; The RGB hybrid white light source is used for imaging of Coomassie blue, silver stained protein glue, and the like.
The C280's user-friendly interface, auto focus, software-controlled light source and filter selection make imaging easier.
Materials and Method
DNA gel
The serially diluted Dongsheng Biotech 100 bp Ladder Plus was separated by gel electrophoresis. Images were acquired on an Azure Biosystems c280 digital imager using a pre-EtBr-stained gel or a gel stained with SYBR Green followed by a UV transilluminator (302 nm) or EPI blue light.
Coomassie blue stained gel
A series of purified transferrins were diluted, diluted with 4-15% Tris-Glycine gel, stained with Coomassie Brilliant Blue dye, and imaged for imaging using an Azure C280 RGB hybrid source.
Chemiluminescence + visible color Marker imaging
The purified transferrin was separated from 500pg-61fg by SDS-PAGE electrophoresis after a series of dilutions. The protein was separated and transferred to a low-fluorescence background membrane using Azure transfer buffer to block and incubate the chicken-anti-transferrin. Primary antibody, rinse, incubate goat-anti-chicken HRP secondary antibody; all dilutions use Azure Chemi Blot Blocking
Buffer. Using Azure Radiance chemiluminescent substrate, imaging was performed using a C280 chemiluminescence module and integrated with a color marker.
Results and discussion
In this application, the Azure Biosystems c280 digital imager is more flexible. The DNA gel results are shown in Figure 1 - A) EB was imaged using UV 302 nm, and B) SYBR Green stained similar gel was imaged with Epi Blue.

Figure 2 shows a representative Coomassie blue stained protein gel. The Azure Biosystems c280 digital imager features a high resolution CCD, automatic image acquisition protocol. The visible imaging mode directly captures the color image, and the contrast is more obvious.

Finally, Figure 3 shows a digital chemiluminescence image combined with a visible color marker, allowing for accurate molecular weight analysis.
